Affinity-purified Amaranthus caudatus (ACL/ACA) is a dimer glycoprotein made up of two subunits. ACL has preferential specificity to oliogsacharrides containing the galactosyl disacharride Galβ3GalNAc. ACL tolerates sialic acid substitution at the 3 position of galactose in the “T” antigen. It does not have specificity for blood groups but does have a higher affinity for type O erythrocytes over B and A.
Alkaline phosphatase (AP) is conjugated to Amaranthus caudatus Lectin (ACL/ACA) to show the binding of ACL/ACA in many applications including Western blotting and ELISA. Alkaline phosphatase is a large protein (140 kDa) that catalyzes the hydrolysis of phosphate groups from a substrate resulting in a colored or fluorescent product. The optimal enzymatic activity of this protein is between pH 8 and 10, and its reaction rate remains linear, improving sensitivity over time.
