DAB (3,3'-diaminobenzidine), anHRP substrate, produces dark brown reaction product withHRP enzyme and is used for both immunohistochemical and Western blotting applications. This chromogen is effective as a single stain or as a second color for double labeling. Since the DAB reaction product is heat-resistant, it can be used in both IHC and ISH labeling applications. Sections developed with DAB can be dehydrated, cleared, and permanently mounted in organic media. The imaging software systems can distinguish DAB from otherHRP or AP enzyme substrates where multiple antigens are co-localized. This substrate can also be used for electron microscopy. The DAB reaction product can be further intensified with DAB Enhancing Solution (see SKU 21760044). Intense™ DAB produces a dark brown reaction product which is 3-4 times more sensitive than the regular DAB substrate produced chromophore. It can be used for both manual and automated staining procedures. ThisKit consists of 100mL of diluent and a concentrated stock solution of purified DAB which can be mixed for series of experiments. Following mixing, the Intense™ DAB solution is stable for up to two weeks when stored at 4‚Äö√†√ªC. Suitable for IHC/ISH and Western blot analysis. This Intense™ DABKit contains all necessary stock solutions in convenient ReadyGo™ dropper bottles format that are easy to use and save time. ThisKit includes DAB ‚àö¬± enhancing solution (Nickel) which can change the color of the reaction product from brown to gray-black and increase sensitivity ofHRP reactions. Sections reacted with DAB/ Ni should be dehydrated, cleared, and permanently mounted in organic media. ThisKit can also be used for Western blot applications. The reagents included in thisKit allows to stain for 300 ‚àö¬± 500 sections and these reagents are stable up to 1 year when stored at 4‚Äö√†√ªC.
