DAB (3,3'-diaminobenzidine), anHRP substrate, produces a dark brown reaction product and this substrate is suitable for both immunohistochemical and Western blot applications. This chromogen is heat-resistance and effective as a single label or as a second color for multiple antigen labeling. Thus, DAB can be used in both IHC or ISH applications and with the aid of imaging systems and appropriate software, the spectral profile of DAB can be distinguished from other enzyme substrates. Sections stained with DAB can also be viewed by darkfield and electron microscopy and dehydrated, cleared, and permanently mounted in organic media. The DAB reaction product can be further intensified with DAB Enhancing Solution (provided). The stock solutions are provided in ReadyGo™ convenient dropper bottles. ThisKit includes nickel, allowing the option of changing the reaction product from brown to gray-black and increasing sensitivity, particularly in blot applications. Slides developed with DAB and Ni should be dehydrated, cleared, and permanently mounted. ThisKit provides all of the necessary reagents to prepare about 300 ml of working solution.
